Effect of Helicobacter pylori HP0355 Gene to Colonize Mice

Abstract
Background/Aims
Helicobacter pylori (H. pylori) is known to be the unique gastric pathogen. The acid resistance of H. pylori has been studied mainly focused on its urease activity. However, independently of the urease activity, it was reported that 10 genes are related to the acid resistance of H. pylori. The purpose of this study is to evaluate the effect of gene HP0355 among these 10 genes to colonize mice. Methods: We first confirmed the presence of HP0355 gene in 12 strains by PCR. Next, the allelic replacement using the antibiotic resistance gene was performed to generate the insertional mutation in gene HP0355. The colonization abilities of the wild type and HP0355 mutant were determined with 6 weeks old C57BL/6 mice. The motilities of these two strains were assessed in soft agar. Results: HP0355 gene was present in all 12 strains. The colonization ability of HP0355 mutant was not different from the wild type. However, HP0355 mutant was better able to colonize mice than the wild type in 1:1 mixed infection. HP0355 mutant had greater motility than the wild type. Conclusions: This study showed that HP0355 gene has acid resistance related to the motility and effects to colonize in gastric mucosa.
